ABSTRACT

BACKGROUND: Studies have shown that Lycium barbarum polysaccharide (LBP) has the functions of anti-aging, nerve protection, anti-fatigue, blood sugar control, anti-oxidation, and anti-tumor. It may have some protective effects against osteoarthritis of the knee, but have been rarely reported. CD151 and matrix metalloproteinase 3 (MMP-3) are two common cytokines for assessing knee osteoarthritis. OBJECTIVE: To observe the effect of LBP on the expression of CD151 and MMP-3 in rabbit osteoarthritis. METHODS: Sixty-four healthy 6-month-old white rabbits were randomly divided into four groups: blank group, model group, LBP group and normal saline group. Animal models of knee osteoarthritis were made using Hulth method in the rabbits except those in the blank group. The rats in the LBP and normal saline groups were fed with normal dose of LBP and normal saline for 4 weeks, and then the articular cartilage tissues were taken from the affected side at 12 weeks after modeling. The morphological changes of the articular cartilage were observed by hematoxylin-eosin staining. The expression levels and spatial distribution of CD151 and MMP-3 in articular cartilage was observed by immunohistochemical staining and western blot. Ethic approval was given by the People’s Hospital of Ningxia Hui Autonomous Region (approval No. 2014-30817). RESULTS AND CONCLUSION: immunohistochemistry staining and western blot results showed that the absorbance values and protein expression of MMP-3 and CD-151 were significantly lower in the LBP group than the normal saline and model groups (P < 0.05). Therefore, the expression of CD151 and MMP-3 in the articular cartilage of osteoarthritis was increased, and LBP could inhibit the expression of CD151 and MMP-3 in osteoarthritis, so as to slow down the occurrence of osteoarthritis.

ABSTRACT

Objective To analyze the relationship between the expression of CD151 and ERK1 and invasion and metastasis of gastric adenocarcinoma,and in order to provide reference for the evaluation of the disease and prognosis. Methods The expression of CD151 and ERK1 in forty-eight cases of gastric adenocarcinoma and adjacent tissues were detected by immunohistochemistry,the relationship between positive expression of CD151 and ERK1 and clinicopathological features was analyzed; the relationship between the expression of CD151 and ERK1 protein and prognosis of the patients was analyzed. Results The positive rates of CD151 and ERK1 in gastric adenocarcinoma tissues were 64. 58%(31/48) and 56. 25%(27/48),which were significantly higher than those of adjacent tissues 10. 42%(5/48) and 16. 67%(8/48),the differences were statistically significant (χ2=30. 044,16. 232,P=0. 000,0. 000);the positive rates of CD151 and ERK1 in TNM stage III±IV,lymph node metastasis,invasion to the serosa and poorly differentiated patients were significantly higher than those in Ⅰ-Ⅱ, non-lymph node metastasis, non-invasion to the serosa and medium and high differentiated patients,the differences were statistically significant (χ2=4. 697,9. 952,7. 288,5. 923,P=0. 030, 0. 002,0. 007,0. 031)(χ2=5. 000,6. 527,9. 257,6. 647,P=0. 025,0. 011,0. 002,0. 036); the patients were followed up to July 2017,the survival rates of CD151 and ERK1 protein positive patients were lower than those of CD151 and ERK1 protein negative patients,the differences were statistically significant (χ2=4. 373,6. 032,P=0. 036,0. 014). Conclusion The expression of CD151 and ERK1 protein in gastric adenocarcinoma tissues is significantly higher, and is closely related to lymph node metastasis, invasion depth, clinical stage and prognosis. It can be used as an important index to evaluate invasion,metastasis and prognosis of patients with gastric adenocarcinoma.

ABSTRACT

Objective: To observe the expressions of CD151 and matrix metalloproteinase-7 (MMP-7) in lung adenocarcinoma tissues and their clinical significance. Methods: The expressions of CD151 and MMP-7 in 149 lung adenocarcinoma tissue samples and 40 adjacent normal tissue samples were detected by immunohistochemistry. The relationship between the expressions of CD151 and MMP-7 and their correlation with the clinicopathological data were analyzed. The disease-free survival (DFS)-related factors were examined. Results: The positive expression rates of CD151 and MMP-7 in lung adenocarcinoma tissues were higher than those in adjacent normal tissues (both P < 0.05). The CD151 expression was associated with smoking, lymph node metastases and TNM stage (all P < 0.05). The MMP-7 expression was associated with lymph node metastases and TNM stage (both P < 0.05). There was a positive correlation between CD151 and MMP-7 expressions in lung adenocarcinoma tissues (r = 0.322, P < 0.05). The DFS of patients with positive expression of CD151 was shorter than that of the patients with negative expression of CD151 (P = 0.005). The DFS of patients with coexpression of CD151 and MMP-7 was shorter than that of the patients with negative expressions of CD151 and MMP-7 (P = 0.004). CD151 was an independent DFS-related factor in patients with lung adenocarcinoma (P < 0.05). Conclusion: The positive expression rates of CD151 and MMP-7 are high in lung adenocarcinoma tissues. The DFS of patients with positive expression of CD151 is relatively short.

ABSTRACT

Objective: To evaluate the clinical indications of CD151 in patients with lung adenocarcinoma with positive expressions of vascular endothelial growth factor A (VEGFA) or vascular endothelial growth factor receptor (VEGFR). Methods: The expressions of VEGFA, VEGFR1, VEGFR2 and CD151 in 116 specimens of patients with lung adenocarcinoma after surgery were detected by immunohistochemistry. The association of CD151 expression with the clinical features of patients with lung adenocarcinoma with positive expression of VEGFA or VEGFR was analyzed. The effect of CD151 expression on the disease-free survival (DFS) of patients with lung adenocarcinoma with positive expression of VEGFA or VEGFR was evaluated. Results: The positive rates of VEGFA, VEGFR1, VEGFR2 and CD151 in lung adenocarcinoma tissues were 69.83% (81/116), 69.83% (81/116), 44.83% (52/116), and 42.24% (49/116), respectively. For patients with lung adenocarcinoma with positive expression of VEGFA, positive expression of CD151 was positively correlated with clinical TNM stage (r = 0.24, P = 0.04) as well as lymph node metastasis (r = 0.26, P = 0.02). CD151 was an independent factor predicting DFS of patients with lung adenocarcinoma with positive expression of VEGFA [hazard ratio (HR) = 1.80, 95% confidence interval (CI): 1.00 - 3.19, P = 0.048]. The median DFS of CD151-positive patients with positive expression of VEGFA was significantly shorter than that of CD151-negative patients (20 vs 34 months, P < 0.05). For patients with lung adenocarcinoma with positive expression of VEGFR1, positive expression of CD151 was positively correlated with TNM stage (r = 0.28, P = 0.01) and lymph node metastasis (r = 0.31, P < 0.01). Moreover, CD151 was an independent factor predicting DFS of patients with lung adenocarcinoma with positive expression of VEGFR2 (HR = 2.10 C95% CI: 1.02 - 4.33, P = 0.044), and the median DFS of CD151-positive patients with positive expression of VEGFR2 was significantly shorter than that of CD151-negative patients (21 vs 42 months, P < 0.05). Conclusion: CD151 is an independent factor predicting DFS of patients with lung adenocarcinoma with positive expression of VEGFA or VEGFR2.

ABSTRACT

Purpose To investigate the expression of the protein of CD151 and MT1-MMP in adenocarcinoma of esophagogastric junc-tion tissues and to explore their relationship with the invasiveness and metastasis of the tumor. Methods CD151 and MT1-MMP pro-tein were detected by immunohistochemical staining respectively in 15 cases of paraneoplastic normal gastric mucosa tissue, 40 cases dysplasia and 172 cases of adenocarcinoma of esophagogastric junction tissues. Results All of the expression level of CD151 and MT1-MMP protein were increasing according to the order of normal-dysplsia-carcinoma. The expression rate of CD151 was 6. 67%, 20. 0% and 78. 3%, while the rate of MT1-MMP with 13. 3%, 22. 5% and 79. 1% in the normal, dysplasia and carcinoma subgroup. The expression rate of each protein were significant difference among the three goroups (P<0. 05). In the adenocarcinoma of esopha-gogastric junction tissues, the expression of them in the subgroup of poorly-differentiated with serosa invasion and lymph nodes metasta-sis was significantly higher than the other subgroup of well-differentiated with non serosa invasion or lymph nodes metastasis ( P <0. 05 ) . There was a positive correlation between the expression of CD151 and MT1-MMP protein in adenocarcinoma of esophagogastric junction tissues ( P<0. 05 ) . Conclusions CD151 and MT1-MMP assuredly and highly express in the adenocarcinoma of esopha-gogastric junction tissues and they have close relationships, which could involved in the invasiveness and metastasis synergistically in this tumor.

ABSTRACT

Objective To investigate the expression and prognostic significance of CD151, c-Met and integrin alpha 3, alpha6 in pancreatic ductal adenocarcinoma (PDAC). Methods The expression of CD151, c-Met and integrin alpha3, alpha6 in 71 patients with PDAC and 10 samples of normal pancreas tissues were detected by immunohistochemistry, and the relationship between the expression of CD151, c-Met and integrin alpha 3, alpha 6 and the clinicopathological features, prognosis of these patients was analyzed. Results The positive expression rates of CD151, c-Met and integrin alpha 3, alpha 6 in PDAC were 81.69% (58/71) , 69.01% (49/71), 69.01% (49/71) and 84.51% (60/71) , and there was no expression in normal pancreas tissues. The expressions of CD151, c-Met were significantly associated with TNM stage and lymph node metastasis (P < 0.05). The expression of CD151 was positively correlated with the expressions of c-Met and integrin alpha3, alpha6 (r =0.583, P =0.000, r = 0.457;P =0.000, r = 0.671 ;P =0.000). Univariate analysis suggested the expression of CD151, c-Met and integrin alpha3, alpha6 was associated with survival (P<0.05). Multivariate analysis suggested the expression of CD151, c-Met was the independent prognostic factor for post-operative survival. Conclusions CD151, c-Met and integrin alpha3, alpha6 play a role in the development, metastasis and prognosis of PDAC, and they might be new markers to predict biological behavior and the prognosis of PDAC patients.

ABSTRACT

The tetraspanin gene, CD151 is involved in various tumour cell progression and metastasis. Its expression is increased in high grade, estrogen receptor negative and c-erbB-2 positive breast cancer. However, the biological function and expression phenotype among different tumour status, estrogen receptor (ER) status, progesterone receptor (PR) status and c-erbB-2 expression in multi-ethnic Malaysian breast cancer patients has not been well investigated. We used quantitative real-time reverse transcriptase polymerase chain reaction (qRT-PCR) to measure the CD151 gene expression in 45 breast cancers. Our preliminary results revealed that CD151 expression is significantly higher in ER positive and PR positive breast cancers at 95% and 99% confidence intervals, respectively. In contrast, there is no significant correlation between CD151 expression and tumour grades or c-erbB-2 status at 95% confidence interval level. Our preliminary findings suggested that CD151 may be involved in the estrogen responsive pathways. CD151 could be a potential prognostic marker and therapeutic target in the treatment of estrogen dependent breast cancer patients.

ABSTRACT

Our previous studies demonstrated that CD151 gene promoted neovascularization in ischemic heart model.To improve the delivery efficacy and target specificity of CD1 51 gene to ischemic heart,we generated an adeno-associated virus (AAV) vector in which CD151 expression was controlled by the myosin light chain (MLC-2v) promoter to achieve the cardiac-specific expression of CD 151 gene in ischemic myocardium and to limit unwanted CD151 expression in extracardiac organs.The function of this vector was examined in rat ischemic myocardium model.The protein expression of CD151 in the ischemic myocardium areas,liver and kidney was confirmed by using Western blot,while the microvessels within ischemic myocardium areas were detected by using immunohistochemistry.The results showed that MLC-2v significantly enhanced the expression of CD151 in ischemic myocardium,but attenuated its expression in other organs.The forced CD151 expression could increase the number of microvessels in the ischemic myocardium.This study demonstrates the AAV-mediated and MLC-2v regulated CD151 gene is highly expressed in the ischemic myocardium and cardiac-specific delivery that is more efficiently targets CD151 to the ischemia myocardium after myocardial infarction.

ABSTRACT

Objective To evaluate the delivery of recombinant adeno-associated virus(rAAV)-mediated CD151 gene in promoting neovascularization and coronary collateralization in swines after myocardial infarction.Method Twenty-six chinese minipigs(clean,provided by breeding pig farm of Huazhong Agricultural University) were randomly divided into four groups:(1)normal control group(n=4):swines without surgery.(2)rAAV-GFP group(n=7):The acute myocardial infarction models were produced in swines by ligating the left anterior descending coronary artery(LAD).The success criteria of the models was that ST-segment elevations in leatds I and aVL maintained for 20 minutes.The rAAV-GFP was injected into the left ventricular anterior wall(divided into 10 points,1×10 11 pfu/point).(3)rAAV-CD151 group(n=7):The operation method was the same as rAAV-GFP group.The rAAV-CD151 was injected into the left ventrieular anterior wall(divided into 10 points).(4) rAAV-antiCD151 group(n=8):The operation method was the same as rAAV-GFP group.The rAAV-antiCD151 was injected into the left ventricular anterior wall(divided into 10 points).Eight weeks after coronary artery liga-tion,the expression of CD151 was measured by western blot.Coronary angiography was done to evaluate collateral circulation of the infarct zone of myocardium.The infurct size was determined by staining with triphenyl-tetrazolium chloride(TTC).Statistical analysis wan carried out by using one-way analysis of variances.Results High level of CD151 protein expression was detected.Coronary angiography showed better collateral circulation in the rAAV-CD151 group.The percentage of infarct size was sinificanly lower in the rAAV-CD151 group (12.82±2.26)% than that in the other two groups,and that was higber in rAAV-anti-CD151 group(32.52±3.47)% than that in the rAAV-GFP group(23.14%±2.83%,both P<0.05).Conelusions CD151 in vivo gene transferred to swines with acute myocardial infarction promotes neovascularization and thereby improves collateral circulation.

ABSTRACT

To investigate the expression of CD151 in human atherosclerosed artery and explore its clinical implications, Western blot and immunohistochemical techniques were used to determine the protein expression of CD151 in arterial tissues with atherosclerosis taken from 36 patients, including 26 cases who received bypass operation for peripheral artery atherosclerosis and 6 cases who died from coronary heart disease. The expression of CD151 in normal artery tissues from 15 healthy organ donators were also measured to serve as control. The results showed that expression of CD151 protein in atherosclerotic arteries was significantly higher than that in normal artery. In ath erosclerotic arteries, CD151 expression was localized in vascular smooth muscle cells (VSMCs) in all tunica media and in partial subintima, while in normal artery, sparse expression was found in tunica media near adventitia. It is concluded that high CD151 protein expression in artery is associated with atherosclerosis and CD151 plays an important role in the atherosclerosis related to VSMC. The expression of CD151 in human atherosclerotic artery depends on the extent of atherosclerotic dam age, it's independent of risk factors.

ABSTRACT

Objective: The signal transduction pathway of phosphatidylinositol-3-kinase(PI3K) may play important roles in promoting angiogenesis induced by growth factors.The involvement of CD151,a member of transmember-4 superfamily,remains unclear.This study aimed to investigate the effects of the recombinant adeno-associated virus mediated CD151(rAAV-CD151) gene delivery on capillary density in myocardial infarction swine and its mechanisms.Methods: We established the swine model of myocardial infarction by coronary artery ligation,and intramuscularly injected rAAV-CD151,rAAV-GFP and rAAV-antiCD151 into the ischemic myocardium.Eight weeks after coronary artery ligation,we detected the expression of CD151,and measured capillary density by immunostaining for the von Willebrand factor.Results: Compared with the control and the rAAV-GFP groups,the rAAV-CD151 group showed a higher CD151 protein expression and capillary density in ischemic myocardium after myocardial infarction(P
